چکیده
A synthetic mimic of carbonic anhydrase shows a lower p K value for the zinc-bound water than natural enzyme. As result, it is able to hydrolyze nonactivated esters at pH 7.

Engineering carbonic anhydrase for highly selective ester hydrolysis
The main part of this thesis describes results from protein engineering experiments, in which the catalytic activity of the enzyme human carbonic anhydrase II (HCAII) is engineered by mutagenesis. This enzyme, which catalyzes the interconversion between CO2 and HCO3in the body, also has the ability to hydrolyze ester bonds. In one project, the specificity of HCAII towards a panel of para-nitrop...
متن کاملInvestigation of solvent effect on the active site energy of Carbonic Anhydrase and Ribonucleotide Reductase
Enzymes catalyze many biological reactions. The rates of chemical reaction in the presence ofenzymes are, in some cases, accelerated more than 10 orders of magnitude relative to thecorresponding rates in solution. In this paper a comparison between optimized structures of two enzyme molecules in aspect ofenergy and dipole moment in different conditions including presence of metallic ion, without...
متن کاملDesign of a carbonic anhydrase IX active-site mimic to screen inhibitors for possible anticancer properties.
Recently, a convincing body of evidence has accumulated suggesting that the overexpression of carbonic anhydrase isozyme IX (CA IX) in some cancers contributes to the acidification of the extracellular matrix, which in turn promotes the growth and metastasis of the tumor. These observations have made CA IX an attractive drug target for the selective treatment of certain cancers. Currently, ther...
متن کاملThe R-state proinsulin and insulin hexamers mimic the carbonic anhydrase active site.
The cobalt(II)-substituted proinsulin and insulin hexamers have been studied in solution via electronic absorption spectroscopy. Hexameric proinsulin is shown to undergo the phenol-induced T6 to R6 conformational transition in a manner analogous to that previously established for insulin. In the absence of coordinating anions, the coordination spheres of the Co(II) ions in the proinsulin and in...
متن کاملMetal binding and catalytic activity in bovine carbonic anhydrase.
Carbonic anhydrase was the earliest known zinc metallo-enzyme (4). It has been isolated from various sources, mainly mammalian red blood cells (see (5)). Although several methods for the preparation of highly active enzyme were described earlier, adequate purification was not achieved nor was the stoichiometry of zinc established (5). Recently, bovine erythrocyte carbonic anhydrase, homogeneous...
